Summary:Ez a munka a Széchenyi-terv: „Hagyományos állatfajták genetikai és gazdasági értékeinek tudományos feltárása“ kutatási program részeként – az ország határain kívül és belül fellelhető sima és fodrostollú lúdállományok felkutatására, a populációk genetikai diverzitásának meghatározására és a közöttük feltételezett genetikai távolság becslésére irányult. A magyar lúd két változata, a fodros- és simatollú lúd, értékes génrezerv fajtánk. Ebben a munkában célom, a magyar lúd fajtához sorolt, de származási dokumentációval nem rendelkező állományok homogenitásának meghatározása, a populációk közötti különbség feltárása volt. Kontrollként emdeni ludat, illetve egy nyári lúd populációt használtam, vizsgálva ezzel a magyar lúd és az emdeni lúd közötti genetikai kapcsolatot is. A kutatómunka során összesen nyolc fodros, öt simatollú és egy emdeni lúd populációból sikerült vérmintához és egy nyári lúd populációból DNS mintához jutnunk. A genetikai variabilitás meghatározásához mikroszatelliteket használtam. A mikroszatelliteket más fajokból kellett adaptálnom (pehely récéből (Somateria mollissima), tőkés récéből (Anas platyrhynchos), kanadai lúdból (Branta canadensis)), mivel mindezidáig sehol a világon nem izoláltak és karakterizáltak domesztikált lúd mikroszatelliteket. A huszonhárom vizsgált mikroszatellitből tizennégyet sikerült a vizsgált fajokban azonosítanom, amiből tíz bizonyult polimorfnak (71%) és négy monomorfnak (29%). A tíz polimorf lokuszon összesen 40 allélt találtam. Az allélszám lokuszonként 2-12 között változott, a heterogenitás 1% és 72%, a PIC értékek pedig 0,12 és 0,83 között alakultak. Az eredményeket a szakirodalmi adatokkal összevetve megállapítottam, hogy az általam adaptált különböző allélszámú és heterozigozitású markerszett házi és nyári lúd populációk genetikai szerkezetének elemzéséhez jól használható. A tíz polimorf mikrosztellittel tanulmányoztam a kísérleti minták genetikai diverzitását. Megállapítottam, hogy három populáció (fodros1, fodros6, fodros8) eltér a Hardy-Weinberg egyensúlytól, ami a csoportokon belül allélveszteségre és a homozigóta egyedek túlsúlyára utal. Három fodrostollú (fodros3, fodros5, fodros7), két simatollú (orosházi, parlagi) és a keresztezett (orosházi x fodros) állományoknál az alacsony allélszám és a viszonylag magas hetrozigozitási értékek alapján, a palacknyak jelenség kezdeti stádiumára következtettem. Két fodrostollú (fodros2, fodros4) és két simatollú magyar (babati1, babati2) lúdcsapatban átlag feletti allélszámot és heterozigozitást mutattam ki, a HWE-tól nem tapasztaltam eltérést. A kontrollként használt emdeni és nyári lúdban szintén átlag feletti allélszámot és a többi populáció viszonyában magas heterozigozitást határoztam meg. Összességében a populációk kifejezetten homogénnek bizonyultak. Legfőbbképpen a fodrostollúak, amik között a különböző színű populációk sem tértek el egymástól szignifikánsan. Mindössze a fodros7 populáció mutatott szignifikáns különbséget a többi magyar – mind fodros, mind simatollú – állományoktól. A simatollú populációk közül az orosházi – babati1 és az orosházi x fodros - babati2 populációk között állapítottam meg szignifikáns eltérést. Különös értéket képviselhet a parlagi jelű csoport, mert, a fodros7 populációhoz hasonlóan, minden más állománytól szignifikánsan különbözött. A Nei-módszerével és a ”közös allélok” (POSA) alapján számszerűsített genetikai távolságok is a fodros7 és parlagi populációk összes többitől való elkülönülését bizonyították. A legnagyobb genetikai távolság értékeket mindkét módszerrel, e két populáció között találtam. Megállapítottam, hogy az 1900-as években történt emdenivel való keresztezés hatása ma is erőteljesen érezhető a magyar állományokban. Erre az szolgál bizonyítékul, hogy a tizenhárom magyar lúd populáció közül, mindössze öt (fodros5 fodros6, fodros7, orosházi, parlagi) tért el szignifikánsan a német fajtától. Ennek a munkának az eredményei jól felhasználhatók a fajtafenntartó tenyésztésben, hiszen a nemzeti kincsé nyilvánított magyar lúd, veszélyeztetett létszámra csökkent, állományának fenntartása elképzelhetetlen széleskörű populációgenetikai ismeretek nélkül. A származási dokumentációval nem rendelkező lúd csoportok homogenitásának meghatározásával, a populációk közötti különbség feltárásával, lehetőség nyílik a fajta fenntartásához szükséges hosszú távú stratégia kidolgozására. A jövőben az általam kidolgozott mikroszatellitszett segítségével, a betenyésztettség változása folyamatosan és rutinszerűen nyomonkövethető. El nem hanyagolható a jelentősége annak, hogy a kidolgozott markerekkel a réghonosult fajtának számító magyar lúd mellett más házilúd fajták molekuláris genetikai jellemzésére is lehetőség nyílik. This essay – as the part of the Széchenyi-plan entitled “Scientfic evaluation of the genetic and economic value of traditional livestock breeds” is aimed at evaluating the plain and curled feathered livestocks, and estimating the genetic distance between the populations as well as defining genetic variability within the populations through molecular genetic markers in-, and around Hungary. There are not many genetic markers to help researchers in relation with poultry species, and this is especially true for goose. Thus, our main objectives were as follows: to adopt microsatellites into domestic goose, to optimalize the microsatellite primers for our examinations and, characterizing the microsatellites found in domestic goose from different species (Common Eider (Somateria mollissima), Mallard duck (Anas platyrhynchos), Canada goose (Branta canadensis)). Our other objectives were to define the genetic variability of livestocks and to estimate the genetic distance with the adaptable microsatellites between the groups, which are claimed to be the curly and plain feathered Hungarian goose, but have no documentation of origin. For the examination we took blood samples from 329 geese, from 14 different places which are not in genetic relation with each other. We used two species for control. One of them was the Greylag goose, the native domestic goose. The other one was the Emden goose, wich was brought to Hungary in the early 1900’s. We examined the genetic relation between the Emden goose and the Hungarian goose as well, because there are more records from the past century regarding the crossing of the Emden breed with the Hungarian goose. From the examined 23 microsatellites we adapted fourteen (61%) to the examined Hungarian-, and Emden goose breeds. Ten out of the fourteen microsatellites were polimorph (71%), and four were monomorph (29%). In the case of the ten locus the number of alleles were between 2–12 in the examined fourteen domestic goose population. Two microsatellites in 21 birds proved to be monomorph from the observed Greylag goose population, the allele number was between 1-10. The expected heterozygote rate was 3%-78%, the observed heterozygote rate was 1-72%. The PIC-rates were between 0.42 and 0.83. In this dissertation, the different variability, adapted microsatellites were perfectly suitable for defining the variability of goose-livestocks diversity and to difference the populations. We have found 40 different alleles on ten locuses in the 15 population all together. The average of the alleles in the population of 15 was 2.6. There were three different populations from the Hardy-Weinberg balance, which indicates overweight homozygote birds inside the population. In three curly feathered, two plain feathered and the crossed populations we can assume that, they are at the early phase of the „bottle-neck” phenomena based on the low rates of the allele numbers and the relatively high rates of the heterozygosis. In two curly feathered and two plain feathered Hungarian populations the allele number and heterozygosis were above the average (2.6). There were no significant differences from the Hardy-Weinberg balance. In the Greylag goose and Emden-goose populations, which are used for control we detected the allele number to be above the average compared to the high heterogeneity of other populations. The curly feathered populations were very homogeneous, only the Curly7 stock was significantly different from the other curly feathered birds. Inside the plain-feathered populations the Parlagi population was different from all others. We found differences between the Babati1- Oroshazi and Babati2- Oroshazi x curly populations as well. We found more differences upon comparing the curly-feathered with the plain-feathered populations. We detected differences between the Oroshazi and four other curly feathered (Curly3, Curly5, Curly6, Curly7) populations. It was a significant difference from the Orosházi x curly the Curly6 and Curly7. The Curly7 was significantly different from the Babati1 the Curly 5, and from Babati 2. The effect of crossing with the Emden breed in the 19001S is even traceable today in the Hungarian populations, because no significant differences were detected between most Hungarian and Emden goose populations. Genetic research of the curly-, and plain feathered versions of Hungarian goose, wich is declared to be our National treasure has never been carried out before. Although the birds decreased to an endangered number, (under 1000) so it is highly unlikely to sustain the livestocks without the much needed population genetic information background. There is an opportunity to work out a strategy for the long run by determining the homogeneity of a population without the documentation of origin. In this dissertation the adopted and characterised microsatellites could be the basis of an easily used marker-set in the future. Repeating the observations frequently with the microsatelites we applied, changes in genetic failure can be detected in ancient and traditional livestocks. Furthermore with the calculated markers there is an opportunity to characterise not only the ancient Hungarian goose, but also some other domestic goose breeds.
