Summary: | Mnoga impresivna dostignuća terapeutskih proteina u liječenju teških bolesti, ne bi bila moguća bez glikoinženjeringa. Terapeutski proteini su sami po sebi kratkog vijeka u organizmu i ograničene su biološke aktivnosti, stoga je nužno uvesti dodatne glikozilacijske mehanizme koji će poboljšati farmakodinamička i farmakokinetička svojstva terapeutskih proteina. U ovom radu su ukratko izloženi osnovni principi glikozilacije proteina u sve tri domene života. Poseban naglasak je stavljen na važnost samog procesa i njegovog utjecaja na stabilnost, strukturu i funkciju (terapeutskih) proteina. Detaljnije su obrađeni načini proizvodnje terapeutskih proteina, problematika glikozilacije humanih proteina u drugim organizmima, te načini postizanja optimalne glikozilacije i dobivanja proteina željene glikoforme. Navedeni su brojni primjeri poboljšanja in vivo biološke aktivnosti proteina glikoinženjeringom, od hiperglikoziliranog analoga humanog rekombinantnog eritropoetina (NESP-a) do terapeutskih protutijela i β-glukocerbrozidaze. Many impressive achievements of therapeutic proteins in treating serious illnesses would not be possible without glycoengineering. Therapeutic proteins have a short serum half-life and limited biological activity. That is why it's necessary to include more glycosylation mechanisms which will consequentially enhance the pharmacokinetic and pharmacodynamic properties of therapeutic proteins. In this paper, the basic glycosylation principles in all domains of life are presented. A special emphasis was given to the importance of the glycosylation process itself on the stability, structure and function of (therapeutic) proteins. In more detail were explained the various methods of synthesis, the problem of glycosylation of human proteins in other organisms and various ways to obtain the wanted glycoform of a therapeutic protein.
