Summary:Poster.-- XVI Congreso Nacional de Virología, Málaga, 6-9 de septiembre de 2022 NK-lysins are a family of antimicrobial peptides conserved in all vertebrate groups that stand out for their antitumor and broad-spectrum antimicrobial activity. The tertiary structure of these peptides shows helical regions stabilized by disulfide bridges that exhibit a lysine-rich facet responsible for its interaction with anionic lipid membranes. In our previous papers, the peptide corresponding to such region in the turbot (Scophthalmus maximus) NK-lysin ortholog (Nkl71-100) showed membrane-disrupting activity against the parasite Philasterides dicentrarchi, as well as inhibition of the pH-dependent fusion step of the entry phase of the spring viremia of carp virus (SVCV). Biophysical data showed that Nkl71-100 preferentially interacts with anionic phosphatidylserine lipid bilayers at low pH conditions. In the present study, the susceptibility of a diverse range of fish viruses to Nkl71-100 has been determined in vitro. In particular, SVCV and tilapia lake virus (TiLV) showed high sensitivity to the Nkl71-100 treatment, while the inhibitory effect on cyprinid herpesvirus 3 (CyHV-3) was only present at higher concentrations and common carp paramyxovirus (CCPV) infectivity was not affected by the peptide.
