Summary: | Halomonasmeridiana(盐单胞菌属)是James等(1990)首次从南极盐湖(盐度从4‰~174‰)中分离出的一株耐盐的盐单胞菌属新种。此属的细菌多生活于极端环境中,既往有关此属细菌的工作仅限于分类和一些极端酶的开发方面,但在基因组学或蛋白质组学方面的研究尚未见报道。本实验首先运用蛋白质组学技术---2-D和质谱技术对H.meridiana的外膜蛋白进行相关研究。由于在数据库中缺少与H.meridiana同属或亲缘关系相近种属的基因组测序结果,限制了依赖于大型数据库的质谱技术的充分运用,为鉴定工作带来困难,难以获得完全可靠的蛋白信息。于是,我们采用了类似真核生物基因组研究的EST.
