Available at http://dx.doi.org/10.1016/j.taap.2008.02.019 The work is part of a larger thematic research initiative, “Airway reactions to biological particles derived from seafood”, and was initiated in 2005 after a study conducted in the Norwegian fish industry revealed increased airway symptoms in fish industry workers. The thesis investigates the ability fish and king crab trypsins have to affect airway epithelial cells and the initiation of intracellular signalling pathways leading to inflammation following receptor activation is examined. Purified salmon, sardine, and king crab trypsins induce the activation and release of inflammatory mediators from human airway epithelial cells. These effects are mediated by activation of protease-activated receptor-2 (PAR-2), a receptor shown to be expressed on many cell types in the airways. Increased production of cytokines from airway epithelial cells is previously shown to be mediated by activation of PAR-2. The work also identifies separate intracellular signalling components that can be linked to inflammation following PAR-2 activation. The issues of the thesis are approached by stimulating epithelial cells that originate from the lower parts of the airways with fish extracts as well as purified trypsins and the effects of these stimulations are analyzed by the use of different molecular biologic methods. The results show that trypsins from fish and king crab can irritate human airway epithelial cells and may have the potential to cause airway inflammation if inhaled in sufficient amounts. Avhandlingen er en del av en større tematisk satsing, "Luftveisreaksjoner ved eksponering for biologiske partikler fra fisk og skalldyr", som ble startet opp i 2005 etter at en undersøkelse viste økt forekomst av luftveisproblemer hos arbeidere i fiskeindustrien sammenlignet med kontrollgrupper. Avhandlingen omhandler evnen trypsin isolert fra fisk og kongekrabbe har til å påvirke overflatecellene i luftveiene våre. Disse enzymenes evne til å igangsette kjedereaksjoner som vil lede til inflammasjon ved å aktivere et bestemt mottakermolekyl (reseptor) på cellenes overflate er undersøkt. I studiene er det avdekket at trypsin fra laks, sardin og krabbe har evnen til å påvirke cellene slik at det dannes signalsubstanser som fra før er kjent at stimulerer en inflammasjonsrespons. Videre er det påvist at disse signalsubstansene dannes som en følge av at protease-aktivert reseptor-2 aktiveres. Denne reseptoren uttrykkes på mange typer celler i luftveiene og er tidligere vist å kunne forårsake inflammasjon ved aktivering. Arbeidet identifiserer også separate komponenter av signalveiene som slås på inne i cellene når denne reseptoren aktiveres og som leder til økt produksjon av inflammasjonsfremmende stoffer (cytokiner). Problemstillingene er belyst ved å stimulere cellekulturer som har opphav fra nedre deler av luftveiene med fiskeekstrakter samt rensede trypsiner og effektene denne stimuleringen har på cellene er analysert ved hjelp av molekylærbiologiske metoder. Resultatene viser at enzymer fra fisk og krabbe kan irritere luftveisceller og ved dette ha potensiale til å skape luftveisinflammasjon dersom de pustes inn i tilstrekkelig grad. |
