A research about the suitable growth culture for S. commune mycelia growth was experimented. One of the aims for this study was to find out the credibility of submerged culture potential in reducing the contamination rate and produce higher mycelia production. Another objective was to investigate the effects of interaction between i media and pH on mycelia growth. The S. commune plant material was collected n Agropark in University Malaysia Kelantan (UMK) Jell Campus and the whole experiment process was done in UMK Tissue Culture laboratory. The collected plant material was cultured into Potato Dextrose Agar (PDA) medium as stock culture and sub-cultured to treated media for 5 days of incubation before data collection was done. The number of contamination for every treated media was taken. Mycelia dry weight was measured and taken after filtered and dried overnight at 70°C. The optimal growth for the mycelia of the fungus was investigated. The contamination rate in conducting. the experiment between two different state of media, liquid and solid media was identified. The media most suitable for the mycelia growth was obtained in solid Mushroom Complete Media (MCM) treated with two nitrogen sources, peptone and yeast extract at pH 5.0, marked at mean dry mycelia weight at 0.405g. Among the treatment conditions, the minimum mycelia growth was found in Potato Dextrose Broth (PDB) at pH 6.0, results in a mean dry mycelia weight at 0.143g. Results indicated that the different pH level would give different effect on different media on mycelia growth. In comparison with the cultured media, the contamination rate in submerged culture (liquid media) was lower than contamination rate in solid artificial media, in which has proven that liquid media would be a more favourable state of media to be used in mycelia culturing and incubating process with lower contamination probability ...
