Das lineare, biologisch abbaubare Polymer Poly(glyceroladipat) (PGA) wurde durch enzymatische Polykondensation unter Verwendung von Lipase B aus Candida antarctica (CAL-B) synthetisiert. Die freien Hydroxylgruppen wurden in unterschiedlichem Maße mit dem entzündungshemmenden Wirkstoff Indomethacin verestert (Steglich Veresterung). Die neuen Polymere eigneten sich für die Formulierung von Nano- und Mikropartikulären Systemen sowie der Herstellung vorgeformter Implantate. Alle Formulierungen zeichneten sich durch kontrollierte Freisetzung ohne Burst Release aus. Die Polymere wiesen reduzierte hämolytische Aktivität sowie verringerte Zelltoxizität im Vergleich zum ungebundenen Indomethacin auf. Die ausgezeichnete Verträglichkeit ausgewählter hochmodifizierter Nanopartikelformulierungen wurde nach Fluoreszenzmarkierung im Mausmodell belegt. The drug is covalently bound via a cleavable linker and remains inactive during its delivery and is activated by specific conditions in the target site. The linear biodegradable polymer poly(glycerol adipate) (PGA) was synthesized by enzymatic polycondensation using lipase B from Candida antarctica (CAL-B). These hydroxyl groups have been esterified to varying degrees with the anti-inflammatory agent indomethacin (Steglich esterification). The new polymers were suitable for the formulation of nano- and microparticulate systems as well as for the production of preformed implants. All formulations showed controlled release without burst release. The polymers had reduced hemolytic activity as well as decreased cell toxicity compared to unbound indomethacin. The excellent tolerability of selected highly modified nanoparticle formulations has been demonstrated after fluorescent labeling in the mouse model.
