Veterinary Proteomics is an evolving scientific field that can provide evidence regarding diagnostic and prognostic biomarkers and set therapeutic targets. Proteomic analysis was performed in several types of biological materials; cerebrospinal fluid (CSF) is a promising source to identify biomarkers for neurological diseases due to the direct contact with the central nervous system (CNS). This prospective study included four dog groups (group A: healthy dogs, group B: dogs with idiopathic epilepsy under antiepileptic medication (AEM), group C: dogs with idiopathic epilepsy without AEM administration, group D: dogs with structural epilepsy). The purpose of the study was to compare the proteomic profile among the four groups. Samples were analyzed by a quantitative Tandem Mass Tags labeling approach using high performance nano-liquid chromatography coupled on-line with a Q-Exactive-Plus mass-spectrometer, able to perform tandem mass spectrometric analysis (MS/MS). Identification and relative quantification were performed using Proteome Discoverer, and data were analyzed using R. Gene ontology terms were analyzed based on Canis lupus familiaris database. Data are available via ProteomeXchange with identifier PXD018893. Eighteen proteins were statistically significant among the four groups (P <0.05). Matrix metalloproteinase 2 (MMP2) and epidermal growth factor (EGF)- containing fibulin-like extracellular matrix protein 2 (EFEMP2) appeared down-regulated whereas haptoglobin (HP) and apolipoprotein A1 (APO-A1) were up-regulated (groups B, D). Tetranectin (CLEC3B) and phosphatidylethanolamine- binding protein 4 (PEBP4) were up-regulated whereas APO-A1 was down-regulated (group C).
