Therefore, the process becomes economically feasible and advantageous due to the repeated usage of enzymes instead of an usage for only one time.In this study, we investigated the immobilization by cross-binding of Thermomyces Lanuginosus (TL 100L) and Candida antarctica lipases. The effects of the pH value of the medium (7.5 – 9.0) and the amount of glutaraldehyde (2-3 mL) in order to immobilize Lipozyme TL 100L and PEG 600 was used as a precipitating agent. In the immobilization process of Candida antarctica, in which ethanol was chosen to precipitate, the pH of the medium was kept at 7.0 – 8.5 and the ratio of enzyme to precipitating agent (1:10 – 1:18) was investigated. Immobilization was performed with usage of Tris-HCl buffer for 24 hours. Immobilization yields were found to be 97% for Lipozyme TL 100L at pH 8.5 and 3 mL of glutaraldehyde. For Candida antartica, the yield was 93% at the same pH value and 1:14 enzyme / precipitating agent ratio. The immobilized enzymes have the potential to be used as biocatalysts. Katalitik proseslerde enzimlerin aktifliğini korumak, ısıl kararlılığını arttırmak, fiziksel dayanımını güçlendirmek ve tekrar tekrar kullanımını sağlamak amacıyla tutuklama işlemi gerçekleştirilir. Böylece enzimlerin tek kullanım yerine defalarca kullanılmasına olanak sağlayarak ekonomik açıdan avantaj elde edilmiş olur. Bu çalışmada, Thermomyces Lanuginosus (TL 100L) ve Candida antarctica lipazlarının çapraz bağlanarak tutuklanması araştırılmıştır. Lipozyme TL 100L tutuklanması için ortam pH’ı (7.5-9) ve glutaraldehit miktarının (2-3 ml) etkileri incelenmiş ve çöktürücü olarak PEG 600 kullanılmıştır. Çöktürücü olarak etanolün kullanıldığı Candida antarctica’nın tutuklanmasında ise, ortam pH’ı 7-8.5 aralığında tutulmuş ve enzim/çöktürücü mol oranının (1:10 - 1:18) etkileri incelenmiştir. Tutuklama işlemleri Tris-HCI tamponu kullanılarak 24 saat süre ile yapılmıştır. Tutuklama verimi, Lipozyme TL 100L için pH 8.5, glutaraldehit miktarı 3 ml koşullarında iken, Candida antarctica için aynı pH değerinde ve enzim /çöktürücü oranı 1:14 değerinde olarak bulunmuştur. Tutuklanan enzimlerin biyokatalizör olarak kullanım potansiyelleri vardır.
