Summary: | This dataset supports the data included in the results chapters of the thesis "Genetic analysis of dynein function in the nervous system and in mitosis". Detailed methodological information is provided in “Chapter 2: Materials and methods” of the thesis. The dataset contains: (1) sixteen movies acquired using an Olympus IX71 inverted microscope with a PerkinElmer UltraVIEW ERS spinning disk system equipped with an Orca Hamamatsu Charge-coupled device (CCD) camera. Movies were acquired with a 60x/1.4 NA PlanApo oil-immersion objective (for movies 3.5) or a 60x/1.2 NA water-immersion objective (for the rest of the movies). Pixel size is 0.11 µm. Image series were processed using Fiji (https://imagej.net/Fiji) or ImageJ (https://imagej.nih.gov/ij/). See “Appendix A” of the thesis for frame rates and information on fluorescence channels for each individual movie. (2) Four Excel spreadsheets with data of identified proteins and unique peptide counts acquired after mass spectrometry of pull-down experiments. Data was visualised using Scaffold 4 (Proteome Software), and exported to Microsoft Excel. See “Appendix A” and “Chapter 5” of the thesis for details on starting material and pull-down methods used in each experiment. |
