Diese Limitierungen können bei kontinuierlicher oder repetitiver Nutzung zu Schwierigkeiten bei der Prozessführung und zu deutlichen Abnahmen der Katalysatorstandzeiten führen. In der vorliegenden Arbeit wurde eine neue Methode zur Stabilisierung von Enzymimmobilisaten entwickelt. Diese ermöglicht eine Erhöhung der mechanischen Stabilität und schützt zugleich vor Enzymleaching, ohne jedoch dadurch drastische Aktivitätseinbußen zu verursachen. Als Beispiel für ein gängiges Enzymimmobilisat diente das kommerzielle Lipasepräparat Novozym 435 (adsorptiv an Polymethylmethacrylat-Träger gebundene Lipase B aus Candida antarctica), das mit 2-Komponenten-Siliconsystemen beschichtet wurde. Bei der Beschichtung füllt ... : Typical problems that often occur when using carrier-bound enzymes such as lipases in technical processes are a successive desorption of enzymes (enzyme-leaching) during the reaction and an insufficient mechanical stability under strong shear stress. These limitations are likely to cause difficulties in technical processes and significantly decrease the catalyst’s lifetime. In this work a novel method to stabilize enzyme preparations was developed. This method allows an increase in mechanical stability and protects the enzyme from desorbing from the carrier without inducing considerable losses in enzymatic activity. Therefore Novozym 435 (Lipase B from Candida antarctica adsorptively bound to a poly(methyl methacrylate) carrier), a widespread commercially available lipase preparation, was herein used as a state-of-the-art biocatalyst for the coating with 2-component silicone systems.
