Siberian sturgeon, Acipenser baeri, spermatozoa : effects of dilution, pH, osmotic pressure, sodium and potassium ions on mobility

International audience The influence of the sperm dilution rate, pH, osmotic pressure and monovalent ion (K(+) Na(+) CI-) concentration on sperm mobility was studied on the Siberian sturgeon, Acipenser baeri. Average values of pH, osmotic pressure, K(+) and Na(+) concentration in the seminal plasma...

Average values of pH, osmotic pressure, K(+) and Na(+) concentration in the seminal plasma were respectively pH 8.1 + or - 0.8, 38 + ou - 3 mOsm/Kg, 2.5 + or - 0.3 and 28 + 7 mM. Titration curve showed a strong buffer capacity of seminal plasma due mainly to presence of phosphate. The best dilution rate in deionized water for observation of motility were about 1/100. Motility occurred in saline solutions of pH 8.2 and osmotic pressure lower than 50 mOsm/Kg. Total inhibition occurred in media of acidic pH and high osmotic pressures (100 mOsm/Kg and more but in this later case, reinitiation of motility was possible after further dilution in deionized water). Potassium at 0.1 mM was strongly, but reversibly inhibiting sperm mobility. Sodium and chloride ions in the range of seminal plasma conditions (20mM) had no effect on the intensity of sperm motility. It was concluded that in physiological conditions, K(+) concentration but not osmolality seemed to be the principal cause of sperm immotility. ...
